Summary
Amantadine is an antiviral drug used for the prophylactic or symptomatic treatment of influenza A, and also as an antiparkinsonian agent to treat extrapyramidal reactions and postherpetic neuralgia. Its antiparkinsonian effect is not fully understood but appears to involve increasing the synthesis and release of dopamine from brain cell nerve endings, possibly inhibiting dopamine uptake, and stimulating the norepinephrine response; it also acts as an NMDA receptor antagonist. Its antiviral mechanism is unrelated, interfering with the viral M2 protein (an ion channel) needed for the viral particle to become uncoated after entering the cell by endocytosis.
