Aliases: 4'-chlorodiazepam, 4-chlorodiazepam
Summary
4'-Chlorodiazepam (Ro5-4864) displays nanomolar affinity for the peripheral benzodiazepine receptor/TSPO located on the outer mitochondrial membrane while showing orders-of-magnitude lower affinity for central GABA-A sites. Low-dose TSPO activation can produce neurosteroidogenesis and mild sedation; higher micromolar concentrations become anxiogenic and pro-convulsant in animals. Median convulsive dose (CDβ β) in mice approximately 42 mg/kg i.p.
Dose Information
Dose basis. Dose ranges are compiled primarily from scattered human anecdotal reports and cautionary community discussions; there are no controlled human PK/PD data. Use a calibrated 0.001 g (1 mg) scale at minimum and strongly consider volumetric dosing to avoid measurement error when working below ~2 mg. Volumetric dosing guidance: dissolve a known mass into a measured volume of an appropriate solvent and dose by volume; see Erowidβs Liquid Measurement Technique for method and pitfalls (solvent choice, evaporation, labeling). (Dose figures and this note are from Drugs.Wiki.)
| ROA | Light | Common | Strong | Heavy |

Add to PsychonautWiki Journal
The PsychonautWiki Journal logging app doesn't include every substance. To track Ro5-4864 there, start adding an ingestion, search for the name, and choose to add it as a custom substance. Paste these values:
Ro5-4864 Source: SubstanceSearch (substancesearch.com) Oral dose (mg): threshold 0.25-0.5 Β· light 0.5-1.5 Β· common 1.5-3 Β· strong 3-5 Β· heavy 5+ (marked seizure risk in animals) Oral duration: onset 0.33-1h Β· peak 1-3h Β· offset 2-3h Β· total 3.33-7h Β· after-effects 12-24h Note: ranges are harm-reduction guidance, not a prescription. Start low, go slow, and test your substances.
Tip: for quicker logging you can also add a Custom Unit in the app, e.g. common dose ~ 2.25 mg (Oral).
The app keeps custom substances simple, so the full dose ranges live in the description text above. Ranges are harm-reduction guidance, not a prescription.
Tolerance
Tolerance Decay
Acute tolerance: develops within a single session β the reset numbers above apply after sustained heavy use, not after one binge. Within-session tachyphylaxis usually resets largely overnight.
Rodent literature shows TSPO desensitization over several days of dosing, but human tolerance dynamics are unknown; treat as potentially habitβforming with partial crossβtolerance only to other TSPO ligands. Data quality is low and largely theoretical/anecdotal.
